Agar used for microbiology is derived from algae, and is a widely used material due to its unique characteristics. As a mostly inert media, Agar is essential for formulating solid and semisolid environment, vital for isolating microorganisms and commonly used as a growth medium for bacteria, fungi and yeast colonies. Agar remains solid at higher temperatures (up to 85°C), making it ideal for incubating cultures without melting and its transparency allows for easy observation and identification of microbial colonies.

Being non-toxic and biocompatible, Agar does not inhibit microbial growth, ensuring a safe and effective medium for microbiological studies. Agar is combined with various nutrients, antibiotics, and indicators to create specific growth conditions tailored to different microorganisms. Agar is used in microbiological research within the pharmaceutical, environmental, cosmetic and food and drink industries.

Efficient PCR-based gene targeting in isolates of the nonconventional yeast Debaryomyces hansenii

by et al. Alhajouj (2023)

The study conducted by et al. Alhajouj (2023) focuses on efficient PCR-based gene targeting in isolates of the nonconventional yeast Debaryomyces hansenii. The research introduces a method that enables high-efficiency gene targeting in wild-type isolates of D. hansenii, addressing the previously reported inefficiency of gene targeting by homologous recombination. This advancement is significant for fundamental and applied biotechnological research involving this yeast species.

In the study, the authors likely utilised Formedium’s Agar powder as a growth medium for the yeast strains during the gene targeting experiments. While the specific details regarding the use of agar are not explicitly mentioned in the reference, it is a common practice in microbiology and molecular biology research to culture yeast on agar plates for various purposes, including gene targeting experiments.

The research by et al. Alhajouj (2023) contributes to the biotechnological potential of D. hansenii by offering a robust method for precise gene targeting, a fundamental tool for genetic manipulation in yeast. This work aligns with the broader context of genome editing and metabolic engineering in non-conventional yeasts, as highlighted in references Cai et al. (2019) and (Löbs et al., 2017). These references emphasize the importance of efficient gene targeting techniques for advancing bioproduction and strain improvement in yeast species.
